5. Touch the radio button corresponding to the desired reaction speed.
The annealing temperature and estimated total run time will change as the speed is changed.
The annealing temperature used in the program will be 4°C below the primer average T

for
the standard speed and 2°C below the primer T
m
average for fast speed. The unadjusted
primer T
m
average will be used for the ultrafast speed.
6. Touch the Next button to review the new protocol in the edit protocol screen.
To edit the protocol, see “Creating, Editing, Naming, and Saving Protocols” on page 11.
7. Touch the Back button to return to the Protocol Autowriter screen.
8. Touch the Save button to save the protocol or the Run button to run the protocol.
Creating a Protocol when the T
a
is Not Known
When the T
a
is unknown, use the instructions to create a protocol with the Protocol AutoWriter.
1. Touch the Protocol AutoWriter button on the home screen.
2. Touch the T
a
Calculator button.
3. Touch the Forward Primer button and enter the primer sequence using the A, T, G, C
buttons. Touch the OK button to confirm.
4. Touch the Reverse Primer button and enter the primer sequence using the A, T, G, C
buttons. Touch the OK button to confirm.
To correct a wrong base entry in the sequence entry pop-up window, press the BACK
button and erase the previously added base.
To erase an entire primer sequence in the T
a
calculator pop-up window, touch the
corresponding clear button.
The Protocol AutoWriter calculates and displays the T
m
of each primer and the average
T
m
and T
a
values (Figure 25).
