Oncethestacksarepositionedinthecassettebase,placethecassettelidonthebase.Thelidis
reversible,butensurethattheelectricalcontactstcloselyintotheslotsinthebase.Presstheliddown
rmlyandturnthedialclockwisetoengagethelidpinsintothelockingslots.
Slidethecassette(withthedialfacingup)intooneoftheTrans-BlotTurboinstrumentbaysuntilitmakes
contactwiththemagneticinterlockinthebackoftheinstrumenttubandyouhearaclick.Thecassette
canbeinsertedintothebayswithorwithoutpowertothesystem.
ThebaysarelabeledA(top)andB(bottom),andcassettescanbeinsertedinterchangeablyintothe
bays.Thereisnorequirementforbothcassettestobeinserted;onebay(eitherAorB)canbeleftempty
whenaprotocolisrun.
Theinstrumentisnowreadytobeginatransferprotocol.Refertothefollowingsectionsfordetailson
transferusingpreprogrammedanduser-denedprotocols.
Note: Atransferforonecassettecanbestartedwhileyouareassemblingasecondcassette.The
secondtransfercanbestartedindependentlyaslongasthesameprotocolisbeingusedforboth.

3.3 Transfer Using RTA Transfer Kits
Trans-BlotTurboRTAtransferkitsprovidethesamereagentsandconsumablesavailableinthetransfer
packs,butinaready-to-assembleformat.Theprotocolforusingthemissimilartothatusedforthe
transferpacks,exceptmembranesandtransferstacksmustbesoakedinTrans-BlotTurbotransfer
bufferbeforeuse.Donotequilibratethegelbeforetransfer.
1. PreparetheTrans-BlotTurboTransferBufferaccordingtoinstructionsonthebottle.
2. Wetandequilibratethemembraneandtransferstacks:
•Nitrocellulosemembrane—immersein30mlof1xtransferbufferandequilibratefor2–3min
•PVDFandLFPVDFmembranes—immersein100%methanolorethanol(usereagentgrade
~85%ormolecularbiologygrade~95%–98%purity)untilthemembraneistranslucent.Transfer
toasoakingtraycontaining30mlof1xtransferbuffer,ensurethemembraneissubmerged,and
equilibrate2–3min
•Transferstacks—
• Midistacks—immersetwostacksseperatedbybluesheetsintwosoakingeachcontaining
50to70mloftransferbufferfor2–3min
• Ministacks—immersetwostacksseperatedbybluesheetsside-by-sideinasoakingtray
containing50to70mloftransferbufferfor2–3min
2.FollowtheproceduredescribedinFigures8-10aboveforassemblingthetransfersandwichesin
transfercassettes,followedbyanaddtionalstep:onceassembled,removeexcesstransferbufferby
invertingthecassettebasewiththeassembledstackcarefullyheldinplaceintoawastecontainer.
Oncetheexcessbufferisremoved,placethecassettelidonthebase,andproceedwiththe
transferstep(3.4).
