Inthe“catchandelute”technique,thereactionmixtureisloadedontothecartridge;theanalytesofinterestareretainedbythesorbent.Awashstepfollowstoremove
additionalreactioncomponentsfromthecartridge.Astrongsolventisusedtoelutetheanalytesfromthecartridge.Sampleconcentrationresultswhenthefinalelution
volume is smaller than the load volume.
Pass Through
Inthepass-throughtechnique,thesorbentdoesnotretainthecompoundsofinterest,butdoesretainsomeclass(es)ofcompoundsandimpurities,thatcanbediscarded
with the column. Fractionation is achieved by the interaction between the impurities in the reaction mixture and the sorbent. No sample enrichment occurs during the
solid-phaseextraction(SPE)step.
Adsorption
Intheadsorptionchromatographytechniqueonareversed-phasesorbent,theneutralcompoundsaremovingdownthechromatographiccolumninthepresenceof
organicsolvent,albeitslowly,forsmall-percentorganicwashes.Loadthereactionmixtureinhighwatercontent
(75%waterforexample),washtoremovepolarcompounds,andthenincreasetheorganiccontentinlargeincrementstoeluteneutrals.Theincreaseisdeterminedby
the hydrophobicity and MW of the compounds.
